The BrdU causes differential contraction along the chromosome and subse quent staining with acridine orange results in fluorescent bands (Du trillaux et al, 1973). A modification of this staining technique is the fluorescent-plus-Giemsa (FPG) technique of Perry and Wolff (1974). This involves staining with 33258 Hoechst, exposing the preparation to exciting light, and then counterstaining with Giemsa. Bands of light and dark Giemsa staining are produced, correlating to the positive and negative fluorescence of the fluorochrome.
